preparation | 2 '-iodouridine (5g,14.1mmol),10% wet palladium carbon (0.1g) and triethylamine (1.8g,18mmol), added to ethanol (100mL), stirred at room temperature for 30min, replaced hydrogen, and maintained the hydrogen pressure in the reaction bottle at 1atm, after stirring at room temperature for 2 hours, TLC showed that the raw materials reacted completely. 2g of diatomite was added. After stirring for 10min, palladium and carbon were removed by suction filtration, the filtrate was concentrated to obtain oil, phosphoric acid (50mL) was added, heated and dissolved, white solid was precipitated after cooling, filtered and washed until almost colorless. After the filter cake was dried, crude product 2'-deoxyuridine -5 '-disodium monophosphate salt was obtained. After recrystallization, filtration and drying were carried out to obtain 2.96g, the yield was 92% and the HPLC purity was 98%. |
Biological activity | 2-deoxyuridine -5 '-disodium monophosphate (dUMP) is the substrate of thymidylate synthase and can be converted into deoxythymidine monophosphate (dTMP). In the methylation step, the inhibitory effect of dUMP on dTMP is the key to control the growth of bacteria and eukaryotic cells. |
